Custom synthesis of aa–tRNAs

Custom  synthesis  of  aa–tRNAs  modified  with  other  dyes  or  probes 

Our custom synthesis service can provide a modified aa-tRNA with the fluorescent dye or other probe of your choice.  This service includes preparing, purifying, and assaying the modified aa-tRNA for activity.  A wide variety of dyes and other probes can be attached to the Lys or Cys side chain in Lys-tRNAamb, Cys-tRNACys, and Lys-tRNALys.  Rather than listing all of the probe and aa-tRNA possibilities, we prefer to first discuss the choice of probe and aa-tRNA with potential customers to ascertain their spectroscopic, biochemical, and other needs.  Since fluorescent dyes of different types and sizes vary greatly in their biological and spectroscopic properties, our experience in using different fluorescent aa-tRNAs may minimize the chances of making a less-than-satisfactory choice.

 
Custom – synthesized  fluorescent  aa-tRNAs  prepared  previously
 
Fluorescent aa-tRNAs containing BODIPY® FL (BOF in our nomenclature), BODIPY® 576/589  (BOP), BODIPY® 558/568 (BOO), or certain other dyes are not sold by tRNA Probes because of trademark restrictions.  However, tRNA Probes will synthesize and purify modified aa-tRNAs containing such dyes if the customer purchases the desired trademarked dye reagent from Invitrogen or other company for use solely in his/her research.  After receiving the reagent from the customer, tRNA Probes will prepare the purified fluorescent aa-tRNA and usually ship it to the customer within 1-2 weeks.
 
The table below shows the cost for the custom synthesis, purification, and assay of various combinations of dyes, tRNAs, and amino acids.  Since the customer purchases and sends the probe reagent to tRNA Probes, the costs below are independent of the cost of probe reagents.
 
Each tube contains 400 pmoles of aa-tRNA that have been dialyzed into 25-30 μl of 1 mM KOAc (pH 5.0), 2 mM Mg(OAc)2.

Custom  synthesis  of  other  modified  aa-tRNAs
 
A variety of probes can be attached either to the side chains of Lys-tRNAamb, Cys-tRNACys, or Lys-tRNALys, and/or to the alpha-amino group of any aa-tRNA to create a peptidyl-tRNA analog.  Such probes include the chemically reactive bromoacetyl group (BrAc), the dinitrophenyl (DNP) affinity probe, and many different fluorescence probes.  If the customer provides the probe reagent and tRNA Probes has experience in working with this reagent, the cost for its custom synthesis will be the same as that of the corresponding aa-tRNA in the above table.  However, if tRNA Probes does not have experience with the probe, the cost for the modified aa-tRNA will be doubled because we will have optimize conditions for the synthesis, purification, and assay of the new modified aa-tRNA using radioactive aa-tRNA.
